Latex Agglutination, Immunodiffusion & Complement Fixation for Antibody Detection

Histoplasmosis is endemic globally, although the most prevalent areas are the central region of North America and Central and South America. Histoplasmosis has been reported in over 60 countries in temperate and tropical climates, including: Africa, Australia, East Asia, India, Indonesia, and Malaysia.
As with other mycotic infections, no single serological test will detect all stages of disease with equal sensitivity. Generally, a combination of tests such as latex and immunodiffusion and complement fixation should be used since it is not usually known when the patient was exposed to the organism. Skin testing for Histoplasma prior to serologic tests should be avoided since a positive skin test may influence serology.

Antibodies in the serum of patients infected with Histoplasma capsulatum will agglutinate the antigen coated latex particles contained in this system. Specimens are usually positive early in the course of primary infection and become negative within the first few months of chronic infection (e.g. the test detect primarily antibodies of the IgM class.)
